Variation of Totipotency from Individual Organ Parts of Hyacinthus orientalis Cultured in vitro at Different Growth Stages



Abstract
The study was carried out to select a useful culture material via test of organogenesis from each part cultured and to get some basic informations on relationship between totipotency and endogeneous substances by that individual part of organs of Hyacinthus orientalis cultivated in disilled water were cultured in vitro. Percentage of organogenesis from all the tissues cultured was higher in the medium containing 1.Omg/l IBA compared with the medium without any auxin.
Organogenesis from bulbscale tissue was better in inner layer of bulbscale than in its outer layer, and in basal part than in distal part. The optimal stage for culture was the first stage, just before planting, and general growth of bulblets was poor.
Organogenesis from leaf tissus was better in leaf tissue inside bulb than in that outside bulb. The :stage which had the best differentiation was identified as the second and third growth stage, in which flower spike was emerged and flowered. Growth of bulblet was pretty good.
Bulblet formation from flower stalk tissue was the best in flower stalk tissue with flower whose stage was at the third stage, flowering period.
Compared with percentage of bulblet formation, number of bulbets productivity of large bulb between tissues cultured, percentage of organogenesis was the highest at leaf tissue, number of bulbets was also the most numerous at leaf tissue but productivity of large bulblet above 100§· was the highest at flower stalk tissue.
Judging from above results, flower stalk tissue with flower which was at flowering period was identified as the best materials for bulblet production.
